Developed robust optical tweezers assays incorporating bioconjugation strategies for tethering DNA-protein complexes in microfluidic environments. Designed single-molecule FRET assays on surface-tethered systems to monitor dynamic conformational changes in real time. Processed large-scale smFRET datasets (10,000 single-molecule trajectories) using MATLAB and analyzed fluorescent intensity and FRET efficiency with Python. Analyzed optical tweezers single-molecule trajectories, trajectory alignment, dwell-time distributions, and pause probabilities using Python and MATLAB.

Investigated telomere protection mechanisms of human shelterin protein complex in vitro. Designed and optimized quantitative FRET titration assays to measure protein-DNA binding kinetics and competition under various experimental conditions. Analyzed titration FRET efficiency, fitted Michaelis-Menten kinetics curves, and compared protein equilibrium constants with Python, MATLAB, and OriginLab.

Developed the ultra-high-precision smFRET method Nanotensioner to resolve Pif1 helicase mechanisms; featured by IOP and published in PRL (2017) and NAR (2016) with broad adoption and multiple citations. Led smFRET assay development including experimental design, surface passivation, and custom acquisition workflows. Built custom analysis algorithms (e.g., FRET transition density step-finding) and statistical models to extract kinetic and mechanistic parameters from single-molecule trajectories. Investigated transcription mechanisms and small-molecule drug inhibition on Mycobacterium tuberculosis RNA polymerase, secured research funding as Principal Investigator through NSFC, and enabled publication in eLife (2025).
